Abstract Eleven Y-chromosomal short tandem repeats (STRs), DYS19, DYS389I, DYS389II, DYS390, DYS391, DYS392, DYS393, DYS385, DYS437, DYS438, DYS439 were typed in DNA samples from Macedonian population (n = 150). Keywords: Y-chromosome; Short tandem repeats; Haplotype; Macedonians ********** Population: One hundred and fifty random male DNA samples from the Macedonian Human DNA Bank (hDNAMKD) [1] were typed for 11 Y-STR markers. DNA samples originated from unrelated males of Macedonian origin, nationality and linguistic affiliation. Individuals are of Christian Orthodox religion and are residents of different geographical regions in the Republic of Macedonia. Extraction: Genomic DNA was isolated from peripheral blood leukocytes by the phenol-chloroform extraction method [2]. The quantity and quality of the genomic DNA isolate was determined by using 260/280 UV spectrophotometer with GeneQuant pro RNA/DNA calculator (Amersham Biosciences). PCR: One multiplex PCR reaction was carried out for each DNA sample using the Power Plex[R] Y System (Promega GmbH, Mannheim) according to the suppliers protocols. Typing: Individual Y-STR alleles were typed using AB 1310 sequencer/software platform. Allele nomenclature according to the Y-STR Haplotype Reference Database, YHRD [3].
